Abstract: The potential for rescuing immature oocytes from the ovaries of females of rare felid species which die or undergo medical ovariohysterectomy was evaluated. Ovaries were recovered from 13 species representing 35 individuals in good-to-poor health. Although the majority of females were 10 yr of age or older and in fair-to-poor health, a total of 846 oocytes were recovered of which 608 (71.9%) were classified as fair-to- excellent quality. One hundred of these oocytes were used for initial maturation classification and as parthogenetic controls. Overall, of the 508 fair-to-excellent quality oocytes placed in culture, 164 (32.3%) matured to metaphase II in vitro. For species in which 3 or more individuals yielded oocytes, mean oocyte maturation rates were as follows: 36.2%, tiger; 27.9% leopard; and 8.3%, cheetah. In vitro insemination of oocytes resulted in fertilization (2 polar bodies, 2 pronuclei, or cleavage) rates of 9.1% to 28.6% (leopard) using homologous fresh spermatozoa and 4.0% (lion) to 40.0% (puma) using homologous frozen-thawed spermatozoa. Inseminations using heterologous (domestic cat) spermatozoa also resulted in fertilized oocytes in the tiger, leopard, snow leopard, puma, serval, and Geoffroy's cat (range in fertilization rate, 5.0% for leopard to 46.2% for puma). Cleaved embryos resulted from the insemination of leopard oocytes with homologous sperm (n = 1 embryo) and puma oocytes with domestic cat sperm (n = 3 embryos). These results demonstrate that immature ovarian oocytes from rare felid species can be stimulated to mature in vitro despite an excision-to-culture interval as long as 36 h.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: Mankial is a sub-valley of the Swat Kohistan. Temperate ecosystem of the valley is intact to a greater extent, which provides habitat to a variety of species of plants, animals and birds. Snow leopard is reported from the valley. To confirm its occurrence, the HUJRA (Holistic Understanding for Justified Research and Action), conducted the study titled “Snow Leopard Survey in Mankial Valley, district Swat, NWFP”. The author provided technical support, while ISLT (The International Snow Leopard Trust) funded the project under its small grants program. The World Wide Fund for Nature-Pakistan (WWF-Pakistan) and the Mankial Community Organization (MCO) facilitated surveys under the project. Surveys revealed that Snow leopard visits parts of the Mankial valley in winter months. Information from the local community shows that Snow leopard remains in the Serai (an off-shoot of the Mankial Valley) from early winter to early spring. Intensive surveys of the prime snow leopard winter habitat in the valley found several snow leopard signs including pugmarks, feces, and scrapes. The study also found occurrence of prey species through indirect evidence though. However, information from the local community confirmed that in the recent past there was a good population of markhor in the valley, which is now reduced to less than 50, mostly due to hunting and habitat disturbance. Hunting is part of the local culture and lifestyle. During winter months hunting pressure is low, as most of the local community migrates to warmer plain areas than Mankial Valley. However, those who live in the area lop oak branches for feeding their livestock and cut trees for burning, in addition to hunting prey species of snow leopard. This has resulted in stunted oak vegetation in most of the lower reaches of the valley and decline of the markhor population.

Abstract: The Ladakh urial (Ovis vignei vignei) is a highly endangered animal (IUCN Red List 2000) listed in the Appendix 1 of CITES and Schedule 1 of the Indian Wildlife Protection Act 1972. Its numbers had been reduced to a few hundred individuals in the 1960s and 70s through hunting for trophies and meat (Fox et al. 1991, Mallon 1983, Chundawat and Qureshi 1999, IUCN Red List 2000). However, with the protection bestowed by the IWPA 1972, and resultant decrease in hunting, the population seems to have shown a marginal increase to about 1000-1500 individuals in its range in Ladakh (Chundawat and Qureshi 1999, IUCN Red List 2000). Although the species had in the past, been able to coexist with the predominantly Buddhist society of Ladakh, the recent increase in the population of both humans and their livestock has placed immense pressures on its habitat (Shackleton 1997, Chundawat and Qureshi 1999, Raghavan and Bhatnagar 2003). This is especially important considering that the Ladakh urial habitat coincides with the areas of maximum human activity in terms of settlements, agriculture, pastoralism and development, in Ladakh (Fox et al. 1991, Chundawat and Qureshi 1999, Raghavan and Bhatnagar 2003). Increased developmental activities such as construction of roads, dams, and military bases in these areas have also increased the access to their habitat. This has consequently made the species more vulnerable to the threats of poaching and habitat destruction (Fox et al. 1991, Chundawat and Qureshi 1999, Raghavan and Bhatnagar 2002). Pressure from increased livestock grazing is one of the major threats faced by the species today (Shackleton 1997, Fox et al. 1991, Mallon 1983, IUCN Red List 2000 Chundawat and Qureshi 1999, Raghavan and Bhatnagar 2003). In the impoverished habitat provided by the Trans-Himalayas, there is great competition for the scarce resources between various animal species surviving here (Fox 1996, Mishra 2001). The presence of livestock intensifies this competition and can either force the species out of its niche (competitive exclusion) by displacing it from that area or resource, or lead to partitioning of resources between the species, spatially or temporally, for coexistence (Begon et al. 1986, Gause 1934).

Abstract: To better understand the biology of snow leopard spermatozoa and to facilitate developing assisted reproduction, a series of studies was conducted to: 1) identify the component(s) of complex culture media responsible for the detrimental effect on sperm survival in vitro, 2) optimize medium for supporting sperm viability, and 3) evaluate sperm capacitation in vitro. Constituents of complex media were added systematically to phosphate-buffered saline (PBS) to isolate the factor(s) influencing snow leopard sperm motility in vitro. Sperm capacitation was also assessed following incubation in PBS with bovine serum albumin (BSA), fetal calf serum (FCS), or heparin. For maintaining sperm motility, there was no benefit (P ? 0.05) to supplementing PBS with low (5%) or high (20%) concentrations of snow leopard serum (SLS) versus FCS or BSA. Likewise, adding supplemental energy substrates (pyruvate, glucose, lactate, or glutamine) did not enhance or hinder (P ? 0.05) sperm motility. However, motility rapidly decreased (P < 0.05) with the addition of NaHCO3 to PBS or Ham's F10 nutrient mixture. Surprisingly, Ham's F10 with no buffering component or with both NaHCO3 and N-Z-hydroxyethylpiperazine-N'-2- ethanesulfonic acid (HEPES) maintained sperm motility at levels similar (P ? 0.05) to PBS. Although sperm motility in all treatments decreased with time, there was a strong inverse relationship (P < 0.01; r = 0.90) between motility and sample pH at 6 hours. Spermatozoa incubated in PBS containing FCS, BSA, or heparin did not undergo the acrosome reaction when exposed to calcium ionophore. In summary, alkaline pH has a profound detrimental effect on snow leopard sperm motility, and capacitation does not occur under conditions that normally promote this event in other felid species. These results clearly demonstrate a high degree of interspecific variation among felids in fundamental sperm function, and they provide evidence for the necessity of basic research when developing assisted reproduction in little-studied nondomestic species.

Abstract: In 2001 the King Mahendra Trust for Nature Conservation (KMTNC), Annapurna Conservation Area (ACAP), Snow Leopard Conservancy (SLC) and WWF-Nepal initiated a collaborative project aimed at enhancing ecotourism in the Manang area, in ways that strengthen benefits to local communities while also protecting the environment and the local culture. Manang is known for its relatively dense snow leopard population, along with supporting good numbers of blue sheep, the endangered cat's principal prey through much of the Himalaya. However, snow leopards periodically kill many livestock, leading to retributive killing by herders along with other associated people-wildlife conflict. In order to encourage the local people to better co-exist with snow leopards and other wildlife, SLC, WWF-Nepal and ACAP agreed to explore ways of providing tourism benefits to local communities as an incentive to protect this rare predator and conserve its alpine habitat. Key in this regard is the possibility of developing locally guided nature treks, and accordingly, this survey was conducted in order to assess existing market opportunities and constraints to such ecotourism enterprise.
