Abstract: Tremendous strides have been made in recent years to broaden our understanding of reproductive processes in nondomestic felid species and further our capacity to use this basic knowledge to control and manipulate reproduction of endangered cats. Much of that progress has culminated from detailed scientific studies conducted in nontraditional laboratory settings, frequently at collaborating zoological parks but also under more primitive conditions, including in the field. A mobile laboratory approach is described, which incorporates a diverse array of disciplines and research techniques. This approach has been extremely useful, especially for conducting gamete characterization and function studies as well as reproductive surveys, and for facilitating the development of assisted reproductive technology. With continuing advances in assisted reproduction in rare felids, more procedures are being conducted primarily as service-related activities, targeted to increase effectiveness of species propagation and population management. It can be a challenge for both investigators and institutional animal care and use committees (IACUCs) to differentiate these service-based procedures from traditional research studies (that require IACUC oversight). For research with rare cat species, multi-institutional collaboration frequently is necessary to gain access to scientifically meaningful numbers of study subjects. Similarly, for service-based efforts, the ability to perform reproductive procedures across institutions under nonstandard laboratory conditions is critical to applying reproductive sciences for managing and preserving threatened cat populations. Reproductive sciences can most effectively assist population management programs (e.g., Species Survival Plans) in addressing conservation priorities if these research and service- related procedures can be conducted “on the road” at distant national and international locales. This mobile laboratory approach has applications beyond endangered species research, notably for other scientific fields (e.g., studies of hereditary disease in domestic cat models) in which bringing the laboratory to the subject is of value.

Abstract: Snow leopards that prey on poor farmers' livestock pose a twofold problem: they endanger farmers' precarious mountain livelihoods as well as the survival of the snow leopard as a unique species since farmers engage in retaliatory killings. Project Snow Leopard (PSL), a recent pilot initiative in Baltistan, involves a partnership between local farmers and private enterprise in the form of an insurance scheme combined with ecotourism activities. Farmers jointly finance the insurance scheme through the payment of premiums per head of livestock they own, while the remaining funds are provided by profits from trekking expeditions focusing on the snow leopard. The insurance scheme is jointly managed by a village management committee and PSL staff. The scheme is structured in such a way that villagers monitor each other and have incentives to avoid cheating the system.

Abstract: Melengestrol acetate (MGA) is the most widely used contraceptive in zoo felids, but the mechanism of contraception and the pathologic effects have not been investigated. For this study, the effects of MGA on folliculogenesis were assessed, and the association of MGA with ovarian lesions was evaluated. Comparisons were made among the histopathologic findings in the ovaries from 88 captive wild felids (representing 15 species) divided into three groups: 37 currently contracepted with MGA, eight previously exposed to MGA, and 43 never contracepted. Ninety-one percent of the felids evaluated had tertiary follicles, and no differences were noted between contracepted and uncontracepted cats. Some MGA-contracepted cats also had corpora lutea indicating recent ovulation. These results indicate that folliculogenesis is not suppressed by current doses of MGA and ovulation occurred in some cats. Therefore, the contraceptive actions of MGA do not occur by suppressing folliculogenesis, and MGA-contracepted felids likely have endogenous estrogens that may confound progestin effects on the uterus. Cystic rete ovarii was the most common pathologic finding, but they were not more prevalent in MGA-contracepted cats. These findings indicate that MGA is not associated with ovarian disease, including ovarian cancer, in contrast to the uterine lesions noted in MGA-treated cats.

Abstract: To better understand the biology of snow leopard spermatozoa and to facilitate developing assisted reproduction, a series of studies was conducted to: 1) identify the component(s) of complex culture media responsible for the detrimental effect on sperm survival in vitro, 2) optimize medium for supporting sperm viability, and 3) evaluate sperm capacitation in vitro. Constituents of complex media were added systematically to phosphate-buffered saline (PBS) to isolate the factor(s) influencing snow leopard sperm motility in vitro. Sperm capacitation was also assessed following incubation in PBS with bovine serum albumin (BSA), fetal calf serum (FCS), or heparin. For maintaining sperm motility, there was no benefit (P ? 0.05) to supplementing PBS with low (5%) or high (20%) concentrations of snow leopard serum (SLS) versus FCS or BSA. Likewise, adding supplemental energy substrates (pyruvate, glucose, lactate, or glutamine) did not enhance or hinder (P ? 0.05) sperm motility. However, motility rapidly decreased (P < 0.05) with the addition of NaHCO3 to PBS or Ham's F10 nutrient mixture. Surprisingly, Ham's F10 with no buffering component or with both NaHCO3 and N-Z-hydroxyethylpiperazine-N'-2- ethanesulfonic acid (HEPES) maintained sperm motility at levels similar (P ? 0.05) to PBS. Although sperm motility in all treatments decreased with time, there was a strong inverse relationship (P < 0.01; r = 0.90) between motility and sample pH at 6 hours. Spermatozoa incubated in PBS containing FCS, BSA, or heparin did not undergo the acrosome reaction when exposed to calcium ionophore. In summary, alkaline pH has a profound detrimental effect on snow leopard sperm motility, and capacitation does not occur under conditions that normally promote this event in other felid species. These results clearly demonstrate a high degree of interspecific variation among felids in fundamental sperm function, and they provide evidence for the necessity of basic research when developing assisted reproduction in little-studied nondomestic species.

Abstract: Zoolites of the Felidae family are known from the Upper Eocene Lower Pliocene in Eurasia, Africa, and North America. Two sub-families are know to inhabit the territory of the USSR and adjacent territories: the extinct sabre-toothed Felidae species Machairodontia and now existing Felidae species. In the USSR the extinct Felidae species were found to exist in Upper Miocene, Upper and Middle Pliocene, and Pleistocene. In Eurasia panthers has been know since early Pliocene. Three species were found in Uzbekistan – the extinct cave lion Felidae sd†l…†… (Goldfuss, 1810), and now existing P…nth†a… tigris, P…nth†a… pardus. The ancient finds and modern habitats are briefly described. Genus Uncia is represented by one species snow leopard or irbis. Probably it appeared in later Pliocene or Pleistocene in the mountain of Central Asia. In Uzbekistan, remains of snow leopard were found in the Samarqand region in the layer of Upper Pleistocene or Holocene. Probably it moved into the area in Pleistocene or the period of glacier removal in the Western Tien Shan mountains, Turkestan, Zeravshan, and Hissar ridges.
