Abstract |
The Pallas’s cat (Otocolobus manul) is one of the most understudied taxa in the Felidae family. The species is currently assessed as being of “Least Concern” in the IUCN Red List, but this assessment is based on incomplete data. Additional ecological and genetic information is necessary for the long-term in situ and ex situ conservation of this species. We identified 29 microsatellite loci with sufficient diversity to enable studies into the individual identification, population structure, and phylogeography of Pallas’s cats. These microsatellites were genotyped on six wild Pallas’s cats from the Tibet Autonomous Region and Mongolia and ten cats from a United States zoo-managed population that originated in Russia and Mongolia. Additionally, we examined diversity in a 91 bp segment of the mitochondrial 12S ribosomal RNA (MT-RNR1) locus and a hypoxia-related gene, endothelial PAS domain protein 1 (EPAS1). Based on the microsatellite and MT-RNR1 loci, we established that the Pallas’s cat displays moderate genetic diversity. Intriguingly, we found that the Pallas’s cats had one unique nonsynonymous substitution in EPAS1 not present in snow leopards (Panthera uncia) or domestic cats (Felis catus). The analysis of the zoo-managed population indicated reduced genetic diversity compared to wild individuals. The genetic information from this study is a valuable resource for future research into and the conservation of the Pallas’s cat. |

Abstract |
During a six-week period in Hemis National Park, Ladakh, India, snow leopards killed 10 sheep and goats and one leopard gained access to a livestock pen and killed many of the animals inside. Dholes also killed sheep and goats, and a wolf killed a young horse. Residents routinely remove snow leopard cubs from their dens to limit future damage by this species. How to deal with the plight of the people living in the area while still protecting the endangered species are major concerns of the International Snow Leopard Trust, which manages Hemis National Park. lgh. |

Abstract |
Pastoralists and their livestock share much of the habitat of the snow leopard (Uncia uncia) across south and central Asia. The levels of livestock predation by the snow leopard and other carnivores are high, and retaliatory killing by the herders is a direct threat to carnivore populations. Depletion of wild prey by poaching and competition from livestock also poses an indirect threat to the region's carnivores. Conservationists working in these underdeveloped areas that face serious economic damage from livestock losses have turned to incentive programs to motivate local communities to protect carnivores. We describe a pilot incentive program in India that aims to offset losses due to livestock predation and to enhance wild prey density by creating livestock-free areas on common land. We also describe how income generation from handicrafts in Mongolia is helping curtail poaching and retaliatory killing of snow leopards. However, initiatives to offset the costs of living with carnivores and to make conservation beneficial to affected people have thus far been small, isolated, and heavily subsidized. Making these initiatives more comprehensive, expanding their coverage, and internalizing their costs are future challenged for the conservation of large carnivores such as the snow leopard. |

Abstract |
To better understand the biology of snow leopard spermatozoa and to facilitate developing assisted reproduction, a series of studies was conducted to: 1) identify the component(s) of complex culture media responsible for the detrimental effect on sperm survival in vitro, 2) optimize medium for supporting sperm viability, and 3) evaluate sperm capacitation in vitro. Constituents of complex media were added systematically to phosphate-buffered saline (PBS) to isolate the factor(s) influencing snow leopard sperm motility in vitro. Sperm capacitation was also assessed following incubation in PBS with bovine serum albumin (BSA), fetal calf serum (FCS), or heparin. For maintaining sperm motility, there was no benefit (P ? 0.05) to supplementing PBS with low (5%) or high (20%) concentrations of snow leopard serum (SLS) versus FCS or BSA. Likewise, adding supplemental energy substrates (pyruvate, glucose, lactate, or glutamine) did not enhance or hinder (P ? 0.05) sperm motility. However, motility rapidly decreased (P < 0.05) with the addition of NaHCO3 to PBS or Ham's F10 nutrient mixture. Surprisingly, Ham's F10 with no buffering component or with both NaHCO3 and N-Z-hydroxyethylpiperazine-N'-2- ethanesulfonic acid (HEPES) maintained sperm motility at levels similar (P ? 0.05) to PBS. Although sperm motility in all treatments decreased with time, there was a strong inverse relationship (P < 0.01; r = 0.90) between motility and sample pH at 6 hours. Spermatozoa incubated in PBS containing FCS, BSA, or heparin did not undergo the acrosome reaction when exposed to calcium ionophore. In summary, alkaline pH has a profound detrimental effect on snow leopard sperm motility, and capacitation does not occur under conditions that normally promote this event in other felid species. These results clearly demonstrate a high degree of interspecific variation among felids in fundamental sperm function, and they provide evidence for the necessity of basic research when developing assisted reproduction in little-studied nondomestic species. |

Abstract |
Livestock depredation has become a significant problem across the snow leopard's (Panthera uncia) range in Central Asia, being most severe in and near protected areas. Such predation, especially incidents of “surplus killing,” in which five to 100 or more sheep and goats are lost in a single night, almost inevitably leads herders to retaliate by killing rare or endangered carnivores like snow leopard, wolf, and lynx. Ironically, such loss can be avoided by making the night-time enclosures predator-proof, improving animal husbandry techniques, educating herders on wildlife conservation and the importance of protecting the natural prey base, and by providing economic incentives like handicrafts skills training and marketing, along with carefully planned ecotourism trekking and guiding. The author explores innovative conservation initiatives in the Himalaya (Ladakh and Tibet) and Mongolia, which also build local capacity, self-reliance, and stewardship for nature using Appreciative Participatory Planning and Action, or APPA, techniques. The most sound conservation investments are those contingent upon establishing direct linkages with biodiversity protection, ensuring co-financing and reciprocal responsibility for project activities, encouraging the full participation of all stakeholders, and assuring regular monitoring and evaluation of the village-based agreements (embodied in Action Plans). |

Abstract |
The globally endangered snow leopard Panthera uncia is sparsely distributed throughout the rugged mountains in Asia.
Its habit of preying on livestock poses a main challenge to management. In the remote Phu valley in northern Nepal, we
obtained reliable information on livestock losses and estimated predator abundance and diet composition from DNA
analysis and prey remains in scats. The annual diet consisted of 42%livestock. Among the wild prey, bharal (blue sheep/
naur) Pseudois nayaur was by far the most common species (92%). Two independent abundance estimates suggested that
there were six snow leopards in the valley during the course of our study. On average, each snow leopard killed about one
livestock individual and two bharal permonth. Predation loss of livestock estimated fromprey remains in scats was 3.9%,
which was in concordance with village records (4.0%). From a total count of bharal, the only large natural prey in the area
and occurring at a density of 8.4 animals/km2 or about half the density of livestock, snow leopards were estimated to
harvest 15.1% of the population annually. This predation rate approaches the natural, inherent recruitment rate of this
species; in Phu the proportion of kids was estimated at 18.4%. High livestock losses have created a hostile attitude against
the snow leopard and mitigation measures are needed. Among innovative management schemes now being implemented
throughout the species’ range, compensation and insurance programmes coupled with other incentive measures are
encouraged, rather than measures to reduce the snow leopard’s access to livestock. In areas like the Phu valley, where the
natural prey base consists mainly of one ungulate species that is already heavily preyed upon, the latter approach, if
implemented, will lead to increased predation on this prey, which over time may suppress numbers of both prey and
predator. |
