Abstract: Most conventional spermatology research involves common mammalian species including livestock, laboratory animals and humans. Yet, there are more than 4500 mammalian species inhabiting the planet for which little is known about basic reproductive biology, including sperm characteristics and function. This information is important, not just as adjunct knowledge, but because the majority of these species are threatened with extinction, largely due to human-induced pressures. The field of conservation is changing rapidly, and global cooperation is emerging among a variety of wildlife enthusiasts, ranging from management authorities of nature reserves to curators of rare zoological collections. Conservation progress depends on systematic, multidisciplinary research first to answer basic questions, with new data then applied to endangered species management plans. The reproductive physiologist is a crucial component of this scheme. Reproduction is the essence of species survival, and enormous effort needs to be directed at these 'untraditional' research species, subspecies and populations. Spermatology research combined with simultaneous efforts in endocrinology, embryology and cryopreservation (among others) can lead to the successful application of assisted reproduction. Examples from this laboratory include an array of wild felid species and a rare cervid and mustelid. Obstacles to success are formidable, including unique species-specificities, diminished genetic diversity and a general lack of resources. Nonetheless, the field offers tremendous opportunities for generating unique knowledge of comparative interest and with conservation utility.

Abstract: Observed a total of 109 copulations in 6 male-female pairs from 4 species of large Felidae. The mean intromission durations were 3.0 sec for Asian leopards (Panthera pardus), 3.3 sec for African leopards (P. pardus), 12.9 sec for snow leopards (Uncia uncia), 2.3 sec for spotted jaguars (P. onca), 3.3 sec for black jaguars (P. onca), and 12.4 sec for Siberian tigers (P. tigris). Behavioral patterns were qualitatively similar across species; all displayed a copulatory pattern with no lock, no intravaginal thrusting, ejaculation on a single insertion, and multiple ejaculations. Whereas domestic cats are reported to assume a neck grip and to tread prior to insertion, these larger Felidae generally did so after intromission had been achieved. Abstract: Eight adult pairs of snow leopards (Panthera uncia) were observed for one to three years in the months December through March to determine the species' social and reproductive characteristics in captivity. To statistically examine the occurrence of behaviors as a function of estrus, the observation weeks were divided into three time blocks: before estrus, estrus, and after estrus. Using percentage of scan samples as an estimate of time spent in various behaviors, 16 behaviors and combined behavior categories were examined for (1) behaviors that differentiated successfully from unsuccessfully breeding pairs, (2) sex differences in behavior, (3) significant correlations between pair members, and (4) behaviors that showed time block effects. The rationale for identifying a behavioral profile of successful breeders in snow leopards was to aid zoos in their captive management programs by increasing their knowledge of the social behavior of this species. By finding correlates to breeding success, informed decisions on whether to change partners after a certain period of time, how to group the cats, and the optimum strategy for a survival plan can be made. Abstract: A syndrome of multiple congenital ocular anomalies in a litter of domestic kittens is described which appears identical to the multiple colobomatous syndrome described in captive Snow Leopards. The lesions varied between kittens in the litter, but ranged from microphthalmos with blindness to mild alterations in the lateral lid margins that resulted in trichiasis. The syndrome of eyelid agenesis in the domestic cat may encompass a broad range of congenital ocular lesions and multiple siblings, but the cause and mechanism of lesion formation is unknown.

Abstract: Blue sheep (Pseudois nayaur) are the main prey of the endangered snow leopard (Panthera uncia) as well as an important game species in Nepal. A knowledge of how blue sheep utilize their habitat is essential for the scientific management of the sheep and for the conservation of the snow leopard, but we only have a limited understanding of this aspect of blue sheep ecology. I studied the habitat use pattern of blue sheep by direct observation in the Anna-purna Conservation Area, Nepal where they occur sympatrically with the snow leopard. The sheep used grassland habitats more frequently during pre-parturition (spring) and post-parturition (autumn) than other habitat types, but scrub and grassland habitats were used equally frequently during the rut (winter). The sheep used smooth undulating slopes of medium steepness (<40 degrees) on southerly aspects within the elevation range of 4,200-4,600 m most frequently in all seasons, and there was no evidence of seasonal migration along the elevation gradient. When not in broken landforms (e.g., cliff, landslides), the sheep maintained proximity (less than or equal to 150 m) to such features suggesting their importance as escape cover (i.e., from predators). The use of habitat components by blue sheep appeared to be related to the distribution of foraging areas and escape cover.

Abstract: A canine beta-casein cDNA was isolated from mammary tissue by polymerase chain reaction (PCR) using degenerate primers. It encodes 250 amino acids protein containing the conserved sequence motif of beta- casein. It showed the highest homology with snow-leopard (Uncia uncia (55-62% identity). It also showed 44-53% identity with human, 33-42%, identity with mouse, 29-37%, identity with rat, 43-53% identity with rabbit, 41-48% identity with pig, 44-51% identity with cattle and 44- 50% identity with sheep. A 1.2-kb mRNA was detected in mammary tissue by Northern blot analysis. Phylogenetic analysis revealed that canine beta-casein formed a branch with lesser panda and snow leopard, which were grouped into carnivore.
