Abstract: Observed a total of 109 copulations in 6 male-female pairs from 4 species of large Felidae. The mean intromission durations were 3.0 sec for Asian leopards (Panthera pardus), 3.3 sec for African leopards (P. pardus), 12.9 sec for snow leopards (Uncia uncia), 2.3 sec for spotted jaguars (P. onca), 3.3 sec for black jaguars (P. onca), and 12.4 sec for Siberian tigers (P. tigris). Behavioral patterns were qualitatively similar across species; all displayed a copulatory pattern with no lock, no intravaginal thrusting, ejaculation on a single insertion, and multiple ejaculations. Whereas domestic cats are reported to assume a neck grip and to tread prior to insertion, these larger Felidae generally did so after intromission had been achieved. Abstract: Noninvasive fecal assays were used to study steroid metabolism and ovarian activity in several felid species. Using the domestic cat (Felis catus) as model, the excretory products of injected [14C]estradiol (E2) and [14C]progesterone (P4) were determined. Within 2 days, 97.0 +/- 0.6% and 96.7 +/- 0.5% of recovered E2 and P4 radioactivity, respectively, was found in feces. E2 was excreted as unconjugated estradiol and estrone (40%) and as a non-enzyme- hydrolyzable conjugate (60%). P4 was excreted primarily as non-enzyme- hydrolyzable, conjugated metabolites (78%) and as unconjugated pregnenolone epimers. A simple method for extracting fecal steroid metabolites optimized extraction efficiencies of the E2 and P4 excretion products (90.1 +/- 0.8% and 87.2 +/- 1.4%, respectively). Analysis of HPLC fractions of extracted fecal samples from the radiolabel-injected domestic cats revealed that E2 immunoreactivity coincided primarily with the unconjugated metabolized [14C]E2 peak, whereas progestogen immunoreactivity coincided with a single conjugated epimer and multiple unconjugated pregnenolone epimers. After HPLC separation, similar immunoreactive E2 and P4 metabolite profiles were observed in the leopard cat (F. bengalensis), cheetah (Acinonyx jubatus), clouded leopard (Neofelis nebulosa), and snow leopard (Panthera uncia). Longitudinal analyses demonstrated that changes in fecal E2 and P4 metabolite concentrations reflected natural or artificially induced ovarian activity. For example, severalfold increases in E2 excretion were associated with overt estrus or exogenous gonadotropin treatment, and elevated fecal P4 metabolite concentrations occurred during pregnant and nonpregnant (pseudopregnant) luteal phases. Although overall concentrations were similar, the duration of elevated fecal P4 metabolites during pseudopregnancy was approximately half that observed during pregnancy. In summary, steroid metabolism mechanisms appear to be conserved among these physically diverse, taxonomically related species. Results indicate that this hormone-monitoring approach will be extremely useful for elucidating the hormonal regulatory mechanism associated with the reproductive cycle, pregnancy, and parturition of intractable and endangered felid species.

Abstract: Nine small cats, including one bobcat (Felis rufus), one Pallas cat (F. manul), one Canada lynx (F. lynx canadensis), two fishing cats (F. viverrina), two margays (F. wiedii), and two sand cats (F. margarita), necropsied between June 1995 and March 1997 had large numbers of gastric spiral bacteria, whereas five large cats, including one African lion (Panthera leo), two snow leopards (P. uncia), one Siberian tiger (P. tigris altaica), and one jaguar (P. onca), necropsied during the same period had none. All of the spiral organisms from the nine small cats were histologically and ultrastructurally similar. Histologically, the spiral bacteria were 5-14 mum long with five to nine coils per organism and were located both extracellularly within gastric glands and surface mucus, and intracellularly in parietal cells. Spiral bacteria in gastric mucosal scrapings from the Canada lynx, one fishing cat, and the two sand cats were gram negative and had corkscrew-like to tumbling motility when viewed with phase contrast microscopy. The bacteria were 0.5-0.7 mum wide, with a periodicity of 0.65-1.1 mum in all cats. Bipolar sheathed flagella were occasionally observed, and no periplasmic fibrils were seen. The bacteria were extracellular in parietal cell canaliculi and intracellular within parietal cells. Culture of mucosal scrapings from the Canada lynx and sand cats was unsuccessful. Based on morphology, motility, and cellular tropism, the bacteria were probably Helicobacter-like organisms. Although the two margays had moderate lymphoplasmacytic gastritis, the other cats lacked or had only mild gastric lymphoid infiltrates, suggesting that these organisms are either commensals or opportunistic pathogens.

Abstract: Nepal's snow leopards (Uncia uncia) are mostly found along the northern border with Tibet (China). The largest populations are in Dolpa, Mugu, Manang, and Myagdi Districts. Potential habitat totals about 30,000 square kilometers. Numbers are estimated at 300-500, but surveys are urgently needed to confirm this rough guess. Like elsewhere, the primary threats center on poaching, depletion of natural prey, livestock depredation and resultant retributive killing of snow leopards by herders, and the lack of public awareness and support for conserving snow leoaprds, especially among local herders.

Abstract: Most conventional spermatology research involves common mammalian species including livestock, laboratory animals and humans. Yet, there are more than 4500 mammalian species inhabiting the planet for which little is known about basic reproductive biology, including sperm characteristics and function. This information is important, not just as adjunct knowledge, but because the majority of these species are threatened with extinction, largely due to human-induced pressures. The field of conservation is changing rapidly, and global cooperation is emerging among a variety of wildlife enthusiasts, ranging from management authorities of nature reserves to curators of rare zoological collections. Conservation progress depends on systematic, multidisciplinary research first to answer basic questions, with new data then applied to endangered species management plans. The reproductive physiologist is a crucial component of this scheme. Reproduction is the essence of species survival, and enormous effort needs to be directed at these 'untraditional' research species, subspecies and populations. Spermatology research combined with simultaneous efforts in endocrinology, embryology and cryopreservation (among others) can lead to the successful application of assisted reproduction. Examples from this laboratory include an array of wild felid species and a rare cervid and mustelid. Obstacles to success are formidable, including unique species-specificities, diminished genetic diversity and a general lack of resources. Nonetheless, the field offers tremendous opportunities for generating unique knowledge of comparative interest and with conservation utility.

Abstract: Modified live feline panleukopenia virus (FPLV) vaccine protected dogs against canine parvovirus (CPV) infection. However, unlike the long- lived (greater than or equal to 20-month) immunity engendered by CPV infection, the response of dogs to living FPLV was variable. Doses of FPLV (snow leopard strain) in excess of 10(5.7) TCID50 were necessary for uniform immunization; smaller inocula resulted in decreased success rates. The duration of immunity, as measured by the persistence of hemagglutination-inhibiting antibody, was related to the magnitude of the initial response to vaccination; dogs with vigorous initial responses resisted oronasal CPV challenge exposure 6 months after vaccination, and hemagglutination-inhibiting antibodies persisted in such dogs for greater than 1 year. Limited replication of FPLV in dogs was demonstrated, but unlike CPV, the feline virus did not spread to contact dogs or cats. Adverse reactions were not associated with living FPLV vaccination, and FPLV did not interfere with simultaneous response to attenuated canine distemper virus.

Abstract: Melengestrol acetate (MGA) is the most widely used contraceptive in zoo felids, but the mechanism of contraception and the pathologic effects have not been investigated. For this study, the effects of MGA on folliculogenesis were assessed, and the association of MGA with ovarian lesions was evaluated. Comparisons were made among the histopathologic findings in the ovaries from 88 captive wild felids (representing 15 species) divided into three groups: 37 currently contracepted with MGA, eight previously exposed to MGA, and 43 never contracepted. Ninety-one percent of the felids evaluated had tertiary follicles, and no differences were noted between contracepted and uncontracepted cats. Some MGA-contracepted cats also had corpora lutea indicating recent ovulation. These results indicate that folliculogenesis is not suppressed by current doses of MGA and ovulation occurred in some cats. Therefore, the contraceptive actions of MGA do not occur by suppressing folliculogenesis, and MGA-contracepted felids likely have endogenous estrogens that may confound progestin effects on the uterus. Cystic rete ovarii was the most common pathologic finding, but they were not more prevalent in MGA-contracepted cats. These findings indicate that MGA is not associated with ovarian disease, including ovarian cancer, in contrast to the uterine lesions noted in MGA-treated cats.

Abstract: Many mammalian herbivores show a temporal diet variation between graminoid-dominated and browse dominated diets. We determined the causes of such a diet shift and its implications for conservation of a medium sized ungulate-the bharal Pseudois nayaur. Past studies show that the bharal diet is dominated by graminoids (>80%) during summer, but the contribution of graminoids declines to about 50% in winter. We tested the predictions generated by two alternative hypotheses explaining the decline: low graminoid availability during winter causes bharal to include browse in their diet; bharal include browse, with relatively higher nutritional quality, in their diet to compensate for the poor quality of graminoids during winter. We measured winter graminoid availability in areas with no livestock grazing, areas with relatively moderate livestock grazing, and those with intense livestock grazing pressures. The chemical composition of plants contributing to the bharal diet was analysed. The bharal diet was quantiWed through signs of feeding on vegetation at feeding locations. Population structures of bharal populations were recorded using a total count method. Graminoid availability was highest in areas without livestock grazing, followed by areas with moderate and intense livestock grazing. The bharal diet was dominated by graminoids (73%) in areas with highest graminoid availability. Graminoid contribution to the bharal diet declined monotonically (50, 36%) with a decline in graminoid availability. Bharal young to female ratio was 3 times higher in areas with high graminoid availability than areas with low graminoid availability. The composition of the bharal winter diet was governed predominantly by the availability of graminoids in the rangelands. Our results suggest that bharal include more browse in their diet during winter due to competition from livestock for graminoids. Since livestock grazing reduces graminoid availability, creation of livestock-free areas is necessary for the conservation of grazing species such as the bharal and its predators including the endangered snow leopard in the Trans-Himalaya.

Abstract: Liver tissues from 126 captive cheetah were evaluated by light microscopy and histochemistry; eight animals were evaluated by electron microscopy. The main hepatic lesion, a vascular lesion resembling veno- occlusive disease (VOD) of the liver and characterized by subendothelial fibrosis and proliferation of smooth muscle-like cells in the central veins, was seen in 60% of the sexually mature cheetah. Although this hepatic vascular lesion was seen in cheetah as young as 1 year of age, the most severe lesions, usually associated with liver failure, were found in cheetah between the ages of 6 and 11. There was no sex predisposition, and in approximately 40% of the VOD cases, liver disease was not suspected clinically or at necropsy. VOD was found in other felidae, especially in the snow leopard. High levels of vitamin A in livers, as well as in diets of the cheetah, could be a contributing factor in the development of VOD in some groups of cheetah.

Abstract: Large carnivores are of vital importance to the stability and integrity of most ecosystems, but recent declines in free-ranging populations have highlighted the potentially devastating effect of infectious diseases on their conservation. We reviewed the literature on infectious diseases of 34 large (maximum body mass of adults >20 kg) terrestrial carnivore species, 18 of which are considered to be threatened in the wild, and examined reports of antibody prevalence (seroprevalence) and cases of infection, mortality and population decline. Of 52 diseases examined, 44% were viral, 31% bacterial and the remainder were protozoal or fungal. Many infections were endemic in carnivores and/or infected multiple taxonomic families, with the majority probably occurring via inhalation or ingestion. Most disease studies consisted of serological surveys for disease antibodies, and antibody detection tended to be widespread implying that exposure to micro-organisms was common. Seroprevalence was higher in tropical than temperate areas, and marginally higher for infections known to occur in multiple carnivore groups. Confirmation of active infection via micro-organism recovery was less common for ursids than other taxonomic groups. Published descriptions of disease-induced population decline or extinction were rare, and most outbreaks were allegedly the result of direct transmission of rabies or canine distemper virus (CDV) from abundant carnivore species to less-common large carnivores. We conclude that the threat of disease epidemics in large carnivores may be serious if otherwise lethal infections are endemic in reservoir hosts and transmitted horizontally among taxa. To prevent or mitigate future population declines, research efforts should be aimed at identifying both the diseases of potential importance to large carnivores and the ecological conditions associated with their spread and severity.

Abstract: It is necessary to look at the big picture when managing biological resources on the QinghaiXizang (Tibetan) plateau. Plateau pikas (Ochotona curzoniae) are poisoned widely across the plateau. Putative reasons for these control measures are that pika populations may reach high densities and correspondingly reduce forage for domestic livestock (yak, sheep, horses), and because they may be responsible for habitat degradation. In contrast, we highlight the important role the plateau pika plays as a keystone species in the Tibetan plateau ecosystem. The plateau pika is a keystone species because it: (i) makes burrows that are the primary homes to a wide variety of small birds and lizards; (ii) creates microhabitat disturbance that results in an increase in plant species richness; (iii) serves as the principal prey for nearly all of the plateau's predator species; (iv) contributes positively to ecosystem-level dynamics. The plateau pika should be managed in concert with other uses of the land to ensure preservation of China's native biodiversity, as well as long-term sustainable use of the pastureland by domestic livestock.

Abstract: In this study, we investigated the cross-reactivity pattern of IgE and IgG4 antibodies to the major feline allergen, Fel d I. We studied the IgE and IgG4 response of 11 cat-allergic patients against Fel d I-like structures in eight members of the Felidae family: ocelot, puma, serval, siberian tiger, lion, jaguar, snow leopard, and caracal. Hair from these “big cats” was collected, extracted, and used in a RAST system and histamine-release test. By means of a RAST-inhibition assay with affinity-purified Fel d I from cat dander, it was established that, in the Felidae species, a Fel d I equivalent is present that reacts with IgE and IgG4 antibodies. We found that all patients had cross-reacting IgE antibodies to seven of the Felidae tested; no IgE antibodies reactive with the caracal were found. Eight of 10 patients with IgG4 antibodies directed to cat dander also had IgG4 antibodies directed to several Felidae species, including the caracal. However, the correlation between the IgE and the IgG4 antibody specificity was low, indicating that, in the case of Fel d I IgE and IgG4, antibodies do not necessarily have the same specificity.

Upon examination, it was found that this nine-yea old, seventy-five pound, wild-trapped animal had sustained considerable dental injuries, including fractures of all maxillary and mandibular anterior teeth. Abstract: In the International Pedigree Book of Snow Leopards 3, Blomqvist and Sten notes (1982) that no information had been recieved on the snow leopard in Mongolia. The present paper sets out to repair that omission by summarising the information in print on snow leopards in Mongolia and giving a brief account of its distribution in the country. This is essentially a review paper and it is hoped that more precise data may be obtained from fieldwork carried out in the future by Mongolian zoologist. The author worked in Mongolia for two years 1975-1977, and during that time collected information on mammals of Mongolia. Information on the snow leopard was obtained from colleagues at the State University of Mongolia; from zoologists and hunters; from herdsmen and local informants from all parts of the country and from three journeys made by the author: to the eastern Gobi Altai; the Khangai mountains, and a 2000 km journey through western Altai. Abstract: In Russia, traditions of track observations and the use of tracking techniques in studying the ecology and behavior of mammals were founded by A.N. Formozov. An analytic review of his data on large carnivorous mammals (tiger, snow leopard, wolf, brown bear, wolverine, and others) is given. A special detailed observation of animals' tracks as a source of information on their life is shown only to start. The efficiency of track observations in various fields of studies, including counting animals, is estimated. The values of day and night distances for various animal species, given in literature, have never been properly substantiated methodically. The tracking method is the most effective in studying the use of the home range by animals, drawing the network of their movements and scent-marking behavior. The hunting behavior of large predators in dense forests is can only be deduced by observing their tracks. In some cases, the use of tracking has a distinct advantage over radio tracking. The main propositions are illustrated by the materials of the author obtained in various Russian regions (in forests of the northern Russian Plain and southern Far east) for 1958-1998.

Abstract: In a litter of three snow leopards, bilateral colobomata of the upper temporal eyelids, bilateral persistent pupillary membranes and a unilateral coloboma of the optic nerve entrance are described as “Multiple Ocular Colobomata” (MOC). The causal pathogenesis of each of the colobomata is discussed comparatively. The colobomata of the eyelids, essential feature of the MOC syndrome in snow leopards, are most probably not of hereditary, but rather of intrauterine infectious viral origin.

Abstract: In 1984, 73 (31/41/1) cubs were born of which 47 (24/23) survived for six months or longer. This gives us a cub mortality of 35.6%. A total of 38 (11/26/1) snow leopards died in captivity in 1984. Five (2/3) specimens were wild caught at the same time in the USSR and then located in the zoos of ALma-Ata, Moscow and Novosibirsk. At the End of 1984, the captive stock consisted of 332 (168/164) snow leopards, an increase of forty animals from the previous year.

Abstract: I studied snow leopards (Panthera uncia) and blue sheep (Pseudois nayaur) in Manang District, Annapurna Conservation Area, Nepal, to estimate numbers and analyze predator-prey interactions. Five to seven adult leopards used the 10-5-km-2 study area, a density of 4.8 to 6.7 leopards/100 km-2. Density of blue sheep was 6.6 10.2 sheep/km-2, and biomass density was 304 kg/km-2. Estimated relative biomass consumed by snow leopards suggested that blue sheep were the most important prey; marmots (Marmota himalayana) also contributed significantly to the diel of snow leopards Snow leopards in Manang were estimated to harvest 9-20% of total biomass and 11-24% of total number of blue sheep annually. Snow leopard: blue sheep ratio was 1:114-1:159 on a weight basis, which was considered sustainable given the importance of small mammals in the leopard's diet and the absence of other competing predators.

Abstract: Home ranges for five radio-tagged snow leopards (Uncia uncia) inhabiting prime habitat in Nepal Himalaya varied in size from 11-37 km2. These solitary felids were crepuscular in activity, and although highly mobile, nearly 90% of all consecutive day movements involved a straight line distance of 2km or less. No seasonal difference in daily movement or home range boundry was detected. While home ranges overlapped substancially, use of common core spaces was temporally seperated, with tagged animals being located 1.9 km or more apart during the smae day. Spatial analysis indicated that 47-55% of use occured within only 6-15% of total home area. The snow leopards shared a common core use area, which was located at a major stream confuence in an area where topography, habitat and prey abundance appeared to be more favorable. A young female used her core area least, a female with two cubs to the greatest extent. the core area was marked significantly more with scrapes, Faeces and other sighn than non-core sites, suggesting that social marking plays an important role in spacing individuals. Snow leopards showed a strong preference for bedding in steep, rocky or broken terrain, on or close to a natural vegetation or landform edge. linear landform features, such as a cliff or major ridgeline, were preferred for travelling and day time resting. This behavior would tend to place a snow leopard close to its preferred prey, blue sheep (Psuedois nayaur), which uses the same habitat at night. Marking was concetrated along commonly travelled routes, particularly river bluffs, cliff ledges and well defined ridgelines bordering stream confluences--features that were most abundant within the core area. Such marking may facilitate mutual avoidance, help maintain the species' solitary social structure, and also enable a relatively high density of snow leopard, especially within high-quality habitat.
