Abstract: The suggestion in the early 20th century that top predators were a necessary component of ecosystems because they hold herbivore populations in check and promote biodiversity was at Wrst accepted and then largely rejected. With the advent of Evolutionary Ecology and a more full appreciation of direct and indirect effects of top predators, this role of top predators is again gaining acceptance. The previous views were predicated upon lethal effects of predators but largely overlooked their non-lethal effects. We suggest that
conceptual advances coupled with an increased use of experiments have convincingly demonstrated that prey experience costs that transcend the obvious cost of death. Prey species use adaptive behaviours to avoid predators, and these behaviours are not cost-free. With predation risk, prey species greatly restrict their use of available habitats and consumption of available food resources. Effects of top predators consequently cascade down to the trophic levels below them. Top predators, the biggies, are thus both the targets of and the means for conservation at the landscape scale.

Abstract: There is recent evidence to suggest that domestic livestock deplete the density and diversity of wild herbivores in the cold deserts of the Trans-Himalaya by imposing resource limitations. To ascertain the degree and nature of threats faced by Himalayan ibex (Capra sibirica) from seven livestock species, we studied their resource use patterns over space, habitat and food dimensions in the pastures of Pin Valley National Park in the Spiti region of the Indian Himalaya. Species diet profiles were obtained by direct observations. We assessed the similarity in habitat use and diets of ibex and livestock using Non-Metric Multidimensional Scaling. We estimated the influence of the spatial distribution of livestock on habitat and diet choice of ibex by examining their co-occurrence patterns in cells overlaid on the pastures. The observed co-occurrence of ibex and livestock in cells was compared with null-models generated through Monte Carlo simulations. The results suggest that goats and sheep impose resource limitations on ibex and exclude them from certain pastures. In the remaining suitable habitat, ibex share forage with horses. Ibex remained relatively unaffected by other livestock such as yaks, donkeys and cattle. However, most livestock removed large amounts of forage from the pastures (nearly 250 kg of dry matter/day by certain species), thereby reducing forage availability for ibex. Pertinent conservation issues are discussed in the light of multiple-use of parks and current socio-economic transitions in the region, which call for integrating social and ecological feedback into management planning.

A few examples are given.

Abstract: Man's hand is turned against the wild cats wherever they occur, often due to the value of their fur, but also because of the danger they sometimes pose to domestic stock and even human beings. All the larger Asian cats are threatened, and on this and the following pages we look at three of them – the Asiatic cheetah, the Siberian tiger, and the snow leopard.

Abstract: Liver tissues from 126 captive cheetah were evaluated by light microscopy and histochemistry; eight animals were evaluated by electron microscopy. The main hepatic lesion, a vascular lesion resembling veno- occlusive disease (VOD) of the liver and characterized by subendothelial fibrosis and proliferation of smooth muscle-like cells in the central veins, was seen in 60% of the sexually mature cheetah. Although this hepatic vascular lesion was seen in cheetah as young as 1 year of age, the most severe lesions, usually associated with liver failure, were found in cheetah between the ages of 6 and 11. There was no sex predisposition, and in approximately 40% of the VOD cases, liver disease was not suspected clinically or at necropsy. VOD was found in other felidae, especially in the snow leopard. High levels of vitamin A in livers, as well as in diets of the cheetah, could be a contributing factor in the development of VOD in some groups of cheetah.

Abstract: Molecular markers that can effectively identify noninvasively collected samples and provide genetic
information are critical for understanding the distribution, status, and ecology of snow leopards (Panthera uncia). However, the low DNA quantity and quality in many
noninvasive samples such as scats makes PCR amplification and genotyping challenging. We therefore designed primers for 9 microsatellites loci previously isolated in the
domestic cat (Felis catus) specifically for snow leopard studies using noninvasive samples. The loci showed moderate levels of variation in two Mongolian snow leopard
populations. Combined with seven other loci that we previously described, they have sufficient variation (He = 0.504, An = 3.6) for individual identification and
population structure analysis. We designed a species species specific PCR assay using cytochrome b for identification of unknown snow leopard samples. These molecular markers
facilitate in depth studies to assess distribution, abundance, population structure, and landscape connectivity of this endangered species.

Abstract: The potential for rescuing immature oocytes from the ovaries of females of rare felid species which die or undergo medical ovariohysterectomy was evaluated. Ovaries were recovered from 13 species representing 35 individuals in good-to-poor health. Although the majority of females were 10 yr of age or older and in fair-to-poor health, a total of 846 oocytes were recovered of which 608 (71.9%) were classified as fair-to- excellent quality. One hundred of these oocytes were used for initial maturation classification and as parthogenetic controls. Overall, of the 508 fair-to-excellent quality oocytes placed in culture, 164 (32.3%) matured to metaphase II in vitro. For species in which 3 or more individuals yielded oocytes, mean oocyte maturation rates were as follows: 36.2%, tiger; 27.9% leopard; and 8.3%, cheetah. In vitro insemination of oocytes resulted in fertilization (2 polar bodies, 2 pronuclei, or cleavage) rates of 9.1% to 28.6% (leopard) using homologous fresh spermatozoa and 4.0% (lion) to 40.0% (puma) using homologous frozen-thawed spermatozoa. Inseminations using heterologous (domestic cat) spermatozoa also resulted in fertilized oocytes in the tiger, leopard, snow leopard, puma, serval, and Geoffroy's cat (range in fertilization rate, 5.0% for leopard to 46.2% for puma). Cleaved embryos resulted from the insemination of leopard oocytes with homologous sperm (n = 1 embryo) and puma oocytes with domestic cat sperm (n = 3 embryos). These results demonstrate that immature ovarian oocytes from rare felid species can be stimulated to mature in vitro despite an excision-to-culture interval as long as 36 h.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: The snow leopard (Panthera uncia Schreber, 1776) population in Russia and Mongolia is situated at the northern edge of the range, where instability of ecological conditions and of prey availability may serve as prerequisites for demographic instability and, consequently, for reducing the genetic diversity. Moreover, this northern area of the species distribution is connected with the western and central parts by only a few small fragments of potential habitats in the Tian-Shan spurs in China and Kazakhstan. Given this structure of the range, the restriction of gene flow between the northern and other regions of snow leopard distribution can be expected. Under these conditions, data on population genetics would be extremely important for assessment of genetic diversity, population structure and gene flow both at regional and large-scale level. To investigate large-scale and fine-grain population structure and levels of genetic diversity we analyzed 108 snow leopards identified from noninvasively collected scat samples from Russia and Mongolia (the northern part of the range) as well as from Kyrgyzstan and Tajikistan (the western part of the range) using panel of eight polymorphic microsatellites. We found low to moderate levels of genetic diversity in the studied populations. Among local habitats, the highest heterozygosity and allelic richness were recorded in Kyrgyzstan (He = 0.66 ± 0.03, Ho = 0.70 ± 0.04, Ar = 3.17) whereas the lowest diversity was found in a periphery subpopulation in Buryatia Republic of Russia (He = 0.41 ± 0.12, Ho = 0.29 ± 0.05, Ar = 2.33). In general, snow leopards from the western range exhibit greater genetic diversity (He = 0.68 ± 0.04, Ho = 0.66 ± 0.03, Ar = 4.95) compared to those from the northern range (He = 0.60 ± 0.06, Ho = 0.49 ± 0.02, Ar = 4.45). In addition, we have identified signs of fragmentation in the northern habitat, which have led to significant genetic divergence between subpopulations in Russia. Multiple analyses of genetic structure support considerable genetic differentiation between the northern and western range parts, which may testify to subspecies subdivision of snow leopards from these regions. The observed patterns of genetic structure are evidence for delineation of several management units within the studied populations, requiring individual approaches for conservation initiatives, particularly related to translocation events. The causes for the revealed patterns of genetic structure and levels of genetic diversity are discussed.

Abstract: The structure of the foramen ovale from 16 species representing 4 carnivore families, the Felidae, Canidae, Ursidae and Hyaenidae, was studied using the scanning electron microscope. The Felidae were represented by 9 domestic cat fetuses (Felis catus), 2 snow leopard neonates (Uncia uncia), an ocelot neonate (Leopardus pardalis), 2 lion neonates (Panthera leo), a panther neonate (Panthera pardus) and 3 tigers (Neofelis tigris), comprising 2 fetuses and a neonate. The Canidae were represented by a golden jackal neonate (Canis aureus), a newborn wolf (Canis lupus), 8 domestic dog fetuses (Canis familiaris), 3 red fox neonates (Vulpes vulpes) and a dhole neonate (Cuon alpinus). The Ursidae were represented by a brown bear neonate (Ursus arctos), a day-old grizzly bear cub (Ursus arctos horribilis), a polar bear neonate (Ursus maritimus), and 2 additional bear fetuses (species unknown). The Hyaenidae were represented by a striped hyaena neonate (Hyaena hyaena). In each species, the foramen ovale, when viewed from the terminal part of the caudal vena cava, had the appearance of a short tunnel. A thin fold of tissue, the developed remains of the embryonic septum primum, extended from the distal end of the caudal vena cava for a variable distance into the lumen of the left atrium and contributed towards the 'tunnel' appearance in all specimens. It constituted a large proportion of the tube, and its distal end was straight-edged. There was fibrous material underlying the endothelium of the flap, the apparent morphology of which suggested that it comprised cardiac muscle.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: Modified live feline panleukopenia virus (FPLV) vaccine protected dogs against canine parvovirus (CPV) infection. However, unlike the long- lived (greater than or equal to 20-month) immunity engendered by CPV infection, the response of dogs to living FPLV was variable. Doses of FPLV (snow leopard strain) in excess of 10(5.7) TCID50 were necessary for uniform immunization; smaller inocula resulted in decreased success rates. The duration of immunity, as measured by the persistence of hemagglutination-inhibiting antibody, was related to the magnitude of the initial response to vaccination; dogs with vigorous initial responses resisted oronasal CPV challenge exposure 6 months after vaccination, and hemagglutination-inhibiting antibodies persisted in such dogs for greater than 1 year. Limited replication of FPLV in dogs was demonstrated, but unlike CPV, the feline virus did not spread to contact dogs or cats. Adverse reactions were not associated with living FPLV vaccination, and FPLV did not interfere with simultaneous response to attenuated canine distemper virus.

Abstract: Non-invasive genetic techniques utilising DNA extracted from faeces hold great promise for felid conservation research. These methods can be used to establish species
distributions, model habitat requirements, analyse diet, estimate abundance and population density, and form the basis for population, landscape and conservation genetic analyses. Due to the elusive nature of most felid species, non-invasive genetic methods have the potential to provide
valuable data that cannot be obtained with traditional observational or capture techniques. Thus, these methods are particularly valuable for research and conservation of endangered
felid species. Here, we review recent studies that use non-invasive faecal genetic techniques to survey or study wild felids; provide an overview of field, laboratory and analysis techniques; and offer suggestions on how future non-invasive genetic studies can be expanded or improved to more effectively support conservation.
