Abstract | The protection of Snow Leopards in the remote and economically disadvantaged Northern Areas of Pakistan needs local people equipped with the skills to gather and present information on the number and range of individual animals in their area. It is important for the success of a conservation campaign that the people living in the area are engaged in the conservation process. Snow Leopards are elusive and range through inhospitable terrain so direct study is difficult. Consequently the major goals for this project were twofold, to gather information on snow leopard distribution in this area and to train local university students and conservation management professionals in the techniques used for locating snow leopards without the need to capture or even see the animals. This project pioneered the use of DNA testing of field samples collected in Pakistan to determine the distribution of snow leopards and to attempt to identify individuals. These were collected in and around that country's most northerly national park, the Kunjurab National Park, which sits on the Pakistan China border. Though the Northern Areas is not a well developed part of Pakistan, it does possess a number of institutions that can work together to strengthen snow leopard conservation. The first of these is a newly established University with students ready to be trained in the skills needed. Secondly WWF-Pakistan has an office in the main town and a state of the art GIS laboratory in Lahore and already works closely with the Forest Department who manage the national park. All three institutions worked together in this project with WWF providing GIS expertise, the FD rangers, and the university students carrying out the laboratory work. In addition in the course of the project the University of the Punjab in Lahore also joined the effort, providing laboratory facilities for the students. As a result of this project maps have been produced showing the location of snow leopards in two areas. Preliminary DNA evidence indicates that there is more than one animal in this relatively small area, but the greatest achievement of this project is the training and experience gained by the local students. For one student this has been life changing. Due to the opportunities provided by this study the student, Nelofar gained significant scientific training and as a consequence she is now working as a lecturer and research officer for the Center for Integrated Mountain Research, New Campus University of the Punjab, Lahore Pakistan |

Abstract | Patterns of mitochondrial restriction fragment length polymorphism (RFLP) variation were used to resolve more recent relationships among the species of the Felidae ocelot lineage, domestic cat lineage, and pantherine lineage. Twenty-five of 28 restriction enzymes revealed site variation in at least 1 of 21 cat species. The ocelot lineage was resolved into three separate sister taxa groups: Geoffroy's cat (Oncifelis geoffroyi) and kodkod (O. guigna), ocelot (Leopardus pardalis) and margay (L. wiedii), and pampas cat (Lynchailurus colocolo) and most of the tigrina samples (Leopardus tigrina). Within the domestic cat lineage, domestic cat (Felis catus), European wild cat (F. silvestris), and African wild cat (F. libyca) formed a monophyletic trichotomy, which was joined with sand cat (F. margarita) to a common ancestor. Jungle cat (F. chaus) and black-footed cat (F. nigripes) mtDNAs diverged earlier than those of the other domestic cat lineage species and are less closely related. Within the pantherine lineage, phylogenetic analysis identified two distinct groups, uniting lion (P. leo) with leopard (P. pardus) and tiger (P. tigris) with snow leopard (P. uncia). | ||||

Abstract | Modern felid species descend from relatively recent (G11 million years ago) divergence and speciation events that produced successful predatory carnivores worldwide but that have confounded taxonomic classifications. A highly resolved molecular phylogeny with divergence dates for all living cat species, derived from autosomal, X-linked, Y-linked, and mitochondrial gene segments (22,789 base pairs) and 16 fossil calibrations define eight principal lineages produced through at least 10 intercontinental migrations facilitated by sea-level fluctuations. A ghost lineage analysis indicates that available felid fossils underestimate (i.e., unrepresented basal branch length) first occurrence by an average of 76%, revealing a low representation of felid lineages in paleontological remains. The phylogenetic performance of distinct gene classes showed that Y-chromosome segments are appreciably more informative than mitochondrial DNA, X-linked, or autosomal genes in resolving the rapid Felidae species radiation. | ||||

Abstract | Livers from 54 snow leopards, 4 days to 23 years old, that had died in 23 US zoos, were evaluated histopathologically to determine if the hepatic fibrosis, which has been noted to be prevalent in this species, was due to chronic active hepatitis from hepadnaviral infection, Ito cell proliferation, or hemosiderosis. Forty-two of 54 snow leopards had subintimal vascular fibrosis with partial or total occlusion of central and sublobular veins (veno-occlusive disease) of unknown origin. All 21 leopards older than 5 years were affected. Four leopards had chronic active hepatitis, and 12 leopards had cholangiohepatitis; but these lesions were not connected anatomically to central and sublobular venous fibrosis. Hepatocellular and Kupffer cell siderosis and Ito cell proliferation were prevalent and often coexisted with perisinusoidal, central, and sublobular venous fibrosis; but fibrosis was present in leopards without siderosis or Ito cell proliferation. The pattern and prevalence of veno-occlusive disease in these leopards was similar to that reported in captive cheetah (Acinonyx jubatus), suggesting that a common extrinsic factor may cause the majority of hepatic disease in these large felid animals in captivity. | ||||

Abstract | Papillomaviruses (PVs) are highly species- and site-specific pathogens of stratified squamous epithelium. Although PV infections in the various Felidae are rarely reported, we identified productive infections in six cat species. PV-induced proliferative skin or mucous membrane lesions were confirmed by immunohistochemical screening for papillomavirus-specific capsid antigens. Seven monoclonal antibodies, each of which reacts with an immunodominant antigenic determinant of the bovine papillomavirus L1 gene product, revealed that feline PV capsid epitopes were conserved to various degrees. This battery of monoclonal antibodies established differential expression patterns among cutaneous and oral PVs of snow leopards and domestic cats, suggesting that they represent distinct viruses. Clinically, the lesions in all species and anatomic sites were locally extensive and frequently multiple. Histologically, the areas of epidermal hyperplasia were flat with a similarity to benign tumors induced by cutaneotropic, carcinogenic PVs in immunosuppressed human patients. Limited restriction endonuclease analyses of viral genomic DNA confirmed the variability among three viral genomes recovered from available frozen tissue. Because most previous PV isolates have been species specific, these studies suggest that at least eight different cat papillomaviruses infect the oral cavity (tentative designations: Asian lion, Panthera leo, P1PV; snow leopard, Panthera uncia, PuPV-1; bobcat, Felis rufus, FrPV; Florida panther, Felis concolor, FcPV; clouded leopard, Neofelis nebulosa, NnPV; and domestic cat, Felis domesticus, FdPV-2) or skin (domestic cat, F. domesticus, FdPV-1; and snow leopard, P. uncia, PuPV-2). | ||||

Abstract | This study reports on two series of cases of Mycobacterium bovis infection in zoo animals. The first was in a captive population of baboons (Papio hamadryas) and the second in a mixed group of wild mammals, including four leopards (Panthera uncia and Panthera pardus) and a sea-lion (Otaria byrona). The isolation and identification of strains of M. bovis confirmed the presence of M. bovis infections in both zoos. The epidemiological study using genetic markers such as the IS6110-based DNA fingerprinting system made it possible to differentiate between M. bovis strains. The M. bovis strains isolated from baboons were shown to contain a single IS6110 copy, as usually do cattle isolates, whereas the M. bovis strains isolated from the other exotic animals presented multiple copies. This finding suggests that the origin of the contamination for the baboons in zoo A could be related to cattle. The origin of the contamination for the leopards and sea-lion in zoo B is more difficult to determine. In conclusion, the authors suggest some recommendations for avoiding outbreaks of tuberculosis infections in zoos. | ||||

Abstract | The purpose of this study was to determine the molecular basis for a relative deficiency in the cat of cytosolic arylamine N- acetyltransferase (NAT), an enzyme family that is important in the metabolism of xenobiotics and that normally consists of at least two related enzymes, NAT1 and NAT2. N-acetyltransferase in feline liver showed high affinity (mean Km = 2.1 microM) for p-aminobenzoic acid, an NAT1 selective substrate in humans and rabbits, but showed a very poor affinity (mean Km > 10 mM) for sulfamethazine, an NAT2 selective substrate in humans and rabbits. Immunoreactive N-acetyltransferase was detected in feline liver, bladder and colon using an NAT1-specific antipeptide antibody, but was not detected in any tissues using an NAT2- specific antibody. Southern blot analysis of genomic DNA demonstrated a single band in domestic cats using each of six restriction digests; single bands were also found on Southern blot analysis of six wild felids. The deduced amino acid sequence of the central portion of feline N-acetyltransferase, obtained by polymerase chain reaction amplification in both domestic cats and seven wild felids (lion, tiger, lynx, snow leopard, bobcat, Asian leopard cat and cheetah), contained three residues, Phe125, Arg127, and Tyr129, which determine NAT1-like substrate specificity in humans. These results support the conclusion that cytosolic arylamine N-acetylation activity is low in the cat because of the presence of a single N-acetyltransferase that has substrate specificity, immunogenicity and sequence characteristics similar to human NAT1, and that the unusual presence of only a single N- acetyltransferase gene appears to be a family wide trait shared by other felids. | ||||

Abstract | The snow leopard (Uncia uncia) is a rare carnivore living above the snow line in central Asia. Using universal primers for the mitochondrial genome control region hypervariable region 1 (HVR1), we isolated a 411-bp fragment of HVR1 and then designed specific primers near each end of this sequence in the conserved regions. These primers were shown to yield good polymerase chain reaction products and to be species specific. Of the 12 snow leopards studied, there were 11 segregating sites and six haplotypes. An identification case of snow leopard carcass (confiscated by the police) proved the primers to be a useful tool for forensic diagnosis in field and population genetics studies. |

Abstract | Polar bears (Ursus maritimus), Eurasian lynx (Lynx lynx) and snow leopards (Panthera uncia) are elusive large carnivores inhabiting snow-covered and remote areas. Their effective conservation and management are challenged by inadequate population information, necessitating development of novel data collection methods. Environmental DNA (eDNA) from snow tracks (footprints in snow) has identified species based on mitochondrial DNA, yet its utility for individual-based analyses remains unsolved due to challenges accessing the nuclear genome. We present a protocol for capturing nuclear eDNA from polar bear, Eurasian lynx and snow leopard snow tracks and verify it through genotyping at a selection of microsatellite markers. We successfully retrieved nuclear eDNA from 87.5% (21/24) of wild polar bear snow tracks, 59.1% (26/44) of wild Eurasian lynx snow tracks, and the single snow leopard sampled. We genotyped over half of all wild polar bear samples (54.2%, 13/24) at five loci, and 11% (9/44) of wild lynx samples and the snow leopard at three loci. Genotyping success from Eurasian lynx snow tracks increased to 24% when tracks were collected by trained rather than untrained personnel. Thirteen wild polar bear samples comprised 11 unique genotypes and two identical genotypes; likely representing 12 individual bears, one of which was sampled twice. Snow tracks show promise for use alongside other non-invasive and conventional methods as a reliable source of nuclear DNA for genetic mark-recapture of elusive and threatened mammals. The detailed protocol we present has utility for broadening end user groups and engaging Indigenous and local communities in species monitoring. | ||||

Abstract | Declining prey populations are widely recognized as a primary threat to snow leopard (Panthera uncia) populations throughout their range. Effective snow leopard conservation will depend upon reliable knowledge of food habits. Unfortunately, past food-habit studies may be biased by inclusion of nontarget species in fecal analysis, potentially misinforming managers about snow leopard prey requirements. Differentiation between snow leopard and sympatric carnivore scat is now cost-effective and reliable using genetics. We used fecal mitochondrial DNA sequencing to identify scat depositors and assessment bias in snow leopard food-habit studies. We compared presumed, via field identification, and genetically confirmed snow leopard scats collected during 2005 and 2012 from 4 sites in Central Asia, using standard forensic microscopy to identify prey species. Field identification success varied across study sites, ranging from 21% to 64% genetically confirmed snow leopard scats. Our results confirm the importance of large ungulate prey for snow leopards. Studies that fail to account for potentially commonplace misidentification of snow leopard scat may mistakenly include a large percentage of scats originating from other carnivores and report inaccurate dietary assessments. Relying on field identification of scats led to overestimation of percent occurrence, biomass, and number of small mammals consumed, but underestimated values of these measures for large ungulates in snow leopard diet. This clarification suggests that the conservation value of secondary prey, such as marmots (Marmota spp.) and other small mammals, may be overstated in the literature; stable snow leopard populations are perhaps more reliant upon large ungulate prey than previously understood. |

Abstract | Non-invasive genetic techniques utilising DNA extracted from faeces hold great promise for felid conservation research. These methods can be used to establish species distributions, model habitat requirements, analyse diet, estimate abundance and population density, and form the basis for population, landscape and conservation genetic analyses. Due to the elusive nature of most felid species, non-invasive genetic methods have the potential to provide valuable data that cannot be obtained with traditional observational or capture techniques. Thus, these methods are particularly valuable for research and conservation of endangered felid species. Here, we review recent studies that use non-invasive faecal genetic techniques to survey or study wild felids; provide an overview of field, laboratory and analysis techniques; and offer suggestions on how future non-invasive genetic studies can be expanded or improved to more effectively support conservation. |

Abstract | Modern felid species descend from relatively recent (<11 million years ago) divergence and speciation events that produced successful predatory carnivores worldwide but that have confounded taxonomic classifications. A highly resolved molecular phylogeny with divergence dates for all living cat species, derived from autosomal, X-linked, Y-linked, and mitochondrial gene segments (22,789 base pairs) and 16 fossil calibrations define eight principal lineages produced through at least 10 intercontinental migrations facilitated by sea-level fluctuations. A ghost lineage analysis indicates that available felid fossils underestimate (i.e., unrepresented basal branch length) first occurrence by an average of 76%, revealing a low representation of felid lineages in paleontological remains. The phylogenetic performance of distinct gene classes showed that Y-chromosome segments are appreciably more informative than mitochondrial DNA, X-linked, or autosomal genes in resolving the rapid Felidae species radiation. |

Abstract | Monitoring Techniques Forest leopards have never been systematically surveyed in African forests, in spite of their potentially vital ecological role as the sole large mammalian predators in these systems. Because leopards are rarely seen in this habitat, and are difficult to survey using the most common techniques for assessing relative abundances of forest mammals, baseline knowledge of leopard ecology and responses to human disturbance in African forests remain largely unknown. This technical handbook sums up the experience gained during a two-year study of leopards by Philipp Henschel in the Lop‚ Reserve in Gabon, Central Africa, in 2001/2002, supplemented by additional experience from carnivore studies conducted by Justina Ray in southwestern Central African Republic and eastern Congo (Zaire) . The main focus of this effort has been to develop a protocol that can be used by fieldworkers across west and central Africa to estimate leopard densities in various forest types. In developing this manual, Henschel tested several indirect methods to assess leopard numbers in both logged and unlogged forests, with the main effort devoted to testing remote photography survey methods developed for tigers by Karanth (e.g., Karanth 1995, Karanth & Nichols 1998; 2000; 2002), and modifying them for the specific conditions characterizing African forest environments. This handbook summarizes the results of the field testing, and provides recommendations for techniques to assess leopard presence/absence, relative abundance, and densities in African forest sites. We briefly review the suitability of various methods for different study objectives and go into particular detail on remote photography survey methodology, adapting previously developed methods and sampling considerations specifically to the African forest environment. Finally, we briefly discuss how camera trapping may be used as a tool to survey other forest mammals. Developing a survey protocol for African leopards is a necessary first step towards a regional assessment and priority setting exercise targeted at forest leopards, similar to those carried out on large carnivores in Asian and South American forests. | ||||
